    Description

In this request, we investigated the comparative risks of thromboembolic stroke, intracranial hemorrhage, gastrointestinal bleeding, and major extracranial bleeding associated with apixaban, dabigatran, and rivaroxaban in patients aged 64 years and younger with non-valvular atrial fibrillation in the Sentinel Distributed Database (SDD).

The objective of this request was to replicate the previous analysis in individuals aged 64 years and younger but utilizing Inverse Probability of Treatment Weighting (IPTW) to adjust for confounding, instead of using Propensity Score Matching (PSM). We conducted pairwise comparisons of apixaban, dabigatran, and rivaroxaban.

This request was divided into three parts, one for each pairwise comparison:

    • Part 1: Compares dabigatran and rivaroxaban use in patients with atrial fibrillation
    • Part 2: Compares apixaban and rivaroxaban use in patients with atrial fibrillation
    • Part 3: Compares apixaban and dabigatran use in patients with atrial fibrillation

The study period includes data from October 19, 2010 to February 28, 2022. We distributed Part 1 on November 15, 2022. We distributed Part 2 on March 20, 2023. We distributed Part 3 on July 7, 2023. All three parts were distributed to five Sentinel Data Partners.

It should be noted that while the study period was identical for each part, the underlying data in each part is slightly different as some Data Partners had refreshed their data in between each distribution date.

The analytic packages associated with this analysis can be found externally in Sentinel's Git Repository at the following locations: Part 1, Part 2, and Part 3. The Git Repository serves as Sentinel's version control tracking system for analytic packages and technical documentation.
